Gene Polymorphisms in NKPD1, APOE, XRCC1, and PEMT and Amnestic Mild Cognitive Impairment: a Small Korean Cohort Study

Abstract

The memory impairment that is typical of amnestic mild cognitive impairment (aMCI) is often accompanied with impairments in executive functioning, especially planning. This is because amnestic MCI is a subtype of mild cognitive impairment. Although it is generally recognized that people with aMCI have planning problems, the brain underpinnings of these deficiencies are still completely unexplored. For this reason, it is of the utmost importance to figure out what causes amyloid-related cognitive impairment (aMCI) and which variables are connected with it in order to make significant progress in the treatment of amyloid-related cognitive impairment.

Methods: We investigated the association between polymorphisms in a number of genes (including NKPD1, APOE, XRCC1 and PEMT) and the risk of developing amnestic mild cognitive impairment (aMCI), as well as the impact of gender differences on aMCI. In our research, there were a total of 263 individuals diagnosed with aMCI as well as 83 non-aMCI (naMCI).

Results: The CC and CT genotypes of the APOE rs429358 polymorphism were related with an elevated risk of Amnestic mild cognitive impairment (aMCI). A statistically significant link between APOE and aMCI was discovered by us. In the group with aMCI, the frequency of the ε4 allele was 22.5%, which was significantly greater than the prevalence in the naMCI group (3.5%). In addition to APOE, we identified three SNPs (rs7946, rs25489 and rs28469095) were associated with an increased risk of aMCI. Of these three SNPs, both the TC and CC genotypes of the XRCC rs7946 polymorphism were linked with an increased risk of age-related macular degeneration in older individuals.

Conclusions: APOE gene rs429358 and XRCC1 gene rs7946 genes may affect the susceptibility to aMCI in a Korean elderly population.
